To evaluate colony growth across experimental libraries, check the control strains (E. coli HB101 and DH5α + pAT64) from Oct 08 and store the plates at 4°C.
Incubated assay selection plates (Inoculated October 06)
Retrieved all plates from the 37°C incubator after overnight incubation.
Visually assessed all plates to look for colonies. Checked control plates side-by-side with experimental libraries to confirm the antibiotic plates were working.
After overnight incubation, the October 08 plates were examined for visible colony growth. Controls came out as expected.
The controls gave the same expected pattern as on Oct 07, and the library plates grew in a similar way. Getting matching results from two separate runs (Oct 06 and Oct 08) makes it much less likely that what we saw came from one bad batch of plates or a contaminated culture.
Because the Oct 06 and Oct 08 plates were both numbered 1–20 (see the note on Oct 08), the two sets were kept apart by date when they went into cold storage.
Pick colonies that grew on IP or MP and re-streak them onto fresh plates to purify them (Oct 14).
